(A) Paper chromatography:

Principle:

The principle of paper chromatography is partition. In paper chromatography, there are two phases: the stationary phase and the mobile phase.

ð The stationary phase is the paper itself (The moisture content of Paper), and

ð the mobile phase is a solvent that moves through the paper.

Compounds that are more soluble in the mobile phase will move faster, while compounds that are more soluble in the stationary phase will move slower. This differential partitioning results in the separation of the compounds into different bands on the paper.

Retention factor is determined using the following formula –

Rf value must be Less than 1

-        Applications:

(i)              To study the process of fermentation

(ii)            To check the purity of phytochemicals

(iii)      To detect adulterants
